تأثیر آلومینیوم بر کاهش رشد و تغییر در ترکیبات دیوارهء سلولهای توتون
Aluminum-induced growth inhibition and changes in cell wall components of tobacco cells.
آلومینیوم از فراوانترین عناصر پوستهء زمین است که میزان آن به 7 درصد کل عناصر میرسد. آلومینیوم یون پیچیدهای است که فرم شیمیایی و عمل بیولوژیک پیچیدهای دارد و در خاکهای خنثی یا خاکهایی با اسیدیته ضعیف به شکل اکسید یا آلومینوسیلیکات غیر قابل حل وجود دارد. با کاهش PH خاک انحلالپذیری این یون افزایش مییابد و از طریق ریشه جذب میشود و بدین ترتیب بر رشد گیاهان تأثیر میگذارد. در واقع، آلومینیوم مهمترین عامل محدود کننده رشد به خصوص رشد ریشه در خاکهای اسیدی شناخته شده است. تقریباً 40-30درصد از زمینهای قابل کشت را خاکهای اسیدی تشکیل میدهند. علاوه بر این اسیدیته خاک به علت مشکلات زیست محیطی نظیر بارانهای اسیدی در حال افزایش است به همین دلیل توجه به سمیت آلومینیوم و تأثیر آن در کاهش بازده تولید محصولات کشاورزی از اهمیت ویژهای برخوردار است. در بسیاری از تحقیقات گذشته تأثیر آلومینیوم در کوتاه مدت نشان داده شده است. همچنین آزمایشهای متعددی نشان داده است که آلومینیوم در ابتدا وارد راس ریشه میشود و غالباً درآپوپلاست جای میگیرد. در واقع آپوپلاست با داشتن ماتریکس پکتینی با شارژ منفی مهمترین محل اتصال آلومینیوم است. این تحقیق به منظور شناسایی هر چه بیشتر تأثیر آلومینیوم بر پلیساکاریدهای دیواره سلولهای توتون انجام گرفت. بدین منظور سلولهای توتون موجود در محیط کشت تعلیقی LS با غلظتهای مختلف آلومینیوم(30 و60 میکرومولار) تیمار داده شدند. پس از 24 ساعت سلولها بر داشت شدندو سپس دیواره سلولی جداسازی شد و اجزا مختلف پلیساکاریدی دیوارهء شامل سلولز، پکتین، همی سلولز A وB استخراج و میزان آنها اندازه گیری شد. همچنین در صد زنده بودن سلولها، میزان جذب آلومینیوم توسط سلولها و میزان رشد آنها بررسی شد. نتایج حاصل از تجزیه و تحلیل دادهها نشان داد که تیمار با آلومینیوم منجر به افزایش همی سلولز A و پکتین در سلولهای توتون شد. امّا اثر کاهشی بر مقدار همی سلولزB مشاهده شد. میزان سلولز نیز در حضور30میکرومولار از آلومینیوم کاهش ولی در حضور60 میکرومولار از آلومینیوم افزایش یافت. همچنین در تیمار با آلومینیوم کاهش رشد و کاهش وزن دیواره مشاهده شد.
Aluminum is one of the most abundant minerals in the soil, comprising approximately 7%. At neutral or weakly acidic PH, Aluminum exists in the form of insoluble aluminosilicate or oxide. However, as the soil becomes more acid, aluminum is solubilized into a phytotoxic form. Aluminum is, therefore a major growth limiting factor especially root growth in acidic soils. It is estimated that approximately 30-40% of arable land in the world is acid soil. Furthermore, the acidity of the soil is gradually increasing as a result of environmental problems including acid rain. Thus, Aluminum toxicity is really an important issue. Under such situations, attention has been paid to improving the agricultural production in acid soil. Inhibition of rapid root growth is reported in some plants. Also, it is suggested that aluminum accumulates primarily and predominantly in the root apoplast where the pectin matrix with its negative changes is a major binding site. This study was undertaken to investigate the effect of aluminum on the amount of cell wall components of tobacco cells. In the present research, cell-cultured tobacco cell (Nicotiana tabacum L. CV. Burley 21) have been used as a plant cell model. After treatment with aluminum for 24 h, cells were harvested. Then, cell wall polysaccharides such as pectin, cellulose, Hemicellulose A and B were isolated from cell wall. Also,cell viability, aluminum absorption by cells and growth were measured. The results indicated that Al-induced increase in the amount of pectin and hemicellulose A. In these cells however, cellulose content reduced with 30µmol but not in the treatment with 60µmol of aluminum. Also loss of cell viability decreased cell growth
