بررسی اثرات کاربرد ماکروسیکلیک دیآمید دیبنزوسولفوکسید بر فعالیت آنزیمهای آنتیاکسیداتیو، بیومارکرهای حاصل از تخریب رادیکالهای اکسیژنی و خصوصیات بافتشناسی کبد و بیضۀ موش نژاد Balb/C
The Study of the Effect of Mecrocyclic Diamid Dibenzo Sulfexide on Antioxidative Enzyme Activities, Oxidative Damage Biomdrkers ad Histological Characteristics of Liver and Testis in Balb/c mice
ماکروسیکلیک دیآمید دیبنزوسولفوکسید، ترکیبی است با ساختار حلقوی-تاجی شکل و دارای گروههای فعال آمیدی، اکسیژنی و سولفوکسید. داخل حلقه آن آب گریز است و میتواند با کاتیونهای سدیم، پتاسیم یا کلسیم پیوند دهد و به دلیل خاصیت چربیدوستی در خارج از حلقه خود میتواند از عرض غشاء عبور کند. پژوهش حاضر با هدف تعیین پاسخ آنزیمهای آنتیاکسیداتیو به اثر غلظتهای متفاوت این ترکیب پس از تزریق درون صفاقی به موشهای نژاد Balb/C انجام گرفت. هم چنین مقادیر بیومارکرهای حاصل از تخریب رادیکالهای اکسیژنی بررسی شدند. نتایج نشان داد که میزان 50 LD این دارو 2500 میکروگرم بر کیلوگرم وزن بدن است. به موشهای بررسی شده دوز50 subLD با غلظت 1500 میکروگرم بر کیلوگرم وزن بدن بهصورت منفرد تزریق شد. بعد از یک هفته بررسی بافتشناسی کبد نشان داد که سلولهای هپاتوسیت، طنابهای هپاتوسیتی، سلولهای کوپفر، سینوزوئیدها و فضای پورتال در مقایسه با گروه شاهد هیچ گونه تغییر معنیداری نیافتند. در حالیکه بافتشناسی بیضهها نشان داد که حجم و وزن بیضه، تعداد سلولهای بینابینی، قطر لولههای منیساز، تعداد سلولهای اسپرماتوگونی نوع A و B، اسپرماتوسیت اولیه، اسپرماتید و اسپرماتوزوئید و سلولهای سرتولی بهطور معنیداری کاهش یافتند. فعالیت آنزیمهای آنتیاکسیداتیو شامل سوپراکسید دیسموتاز، کاتالاز، گلوتاتیون پراکسیداز در کبد و بیضه در یک روند وابسته به غلظت رو به افزایش گذاشتند در صورتی که این افزایش برای بیومارکرهای تخریب اکسیداتیو یعنی مالون دیآلدئید و دیتیروزین فقط در بافت بیضه و در خون محیطی مشاهده شد. بنا بر این با توجه به پژوهش حاضر نتیجهگیری میشود که ترکیب مورد استفاده دارای اثرات سمی بوده است. بعد از یک هفته از کاربرد آن در زیر غلظت 50 LD، بافت بیضه به نحو شاخص تحت تأثیر قرار گرفت، ولی بافت کبد احتمالاً به دلیل قابلیتهای متابولیک زیاد در متابولیسم مواد و توان بیشتر فعالیت آنتیاکسیدانی، اثرات نامطلوب بافتی نشان نداد.
Dibenzo sulfoxide macrocyclic diamides possess a hydrophilic cavity surrounded by hydrophobic ring, which enable them to diffuse from cell membrane and interfere with different living systems including ion channels. This study evaluated cytotoxicity effects of this compound in a range of different doses on Balb/C mouse via inter peritoneal injection. Results showed that LD50 concentration was at 2500 µg/Kg.bodyweight. Histological evaluation of animals livers after one-week treatment to 1500 µg/Kg of this compound revealed insignificant changes in hepatocyte cords, portal area, sinusoids and cells of coopfer and hepatocytes with respect to control animals. On the other hand, Histological evaluation of animals’ testis indicated significant decrease in weight, seminiferous tubule diameter and volume of testis and also in the cell number of spermatogonia (A and B), primary spermatocytes, spermatides, spermatozoids and sertoli. In this condition of exposure, antioxidative enzyme activities of peripheral blood, liver and testis increased in a dose dependent manner and reached the highest and significant levels at 1500 µg/Kg. However, the increase in oxidative damage biomarkers including malondialdehyde (MDA) and dityrosine were considerable for peripheral blood and testis. Accordingly, the studied compound had toxic effect and could affected testis when the animals were treated with chemical agent subLD50 for one week. The liver didn’t show toxicity effect of this compound even at 1500 µg/Kg exposure for one week. It could be attributed to high metabolism potential and marked antioxidative activities of this tissue.
