Abstract

Fish oil in the diet of fish is constantly at the risk of oxidation. In this study, the effects of feeding sturgeon hybrid (Huso huso ♂ ×Acipenser ruthenus ♀) with different levels of oxidized fish oil (OFO) on thyroid hormones were investigated. Three experimental diets were made by replacing 0 (control), 50 and 100% OFO were made. Ninety hybrid sturgeon, with the initial weights of 212.6 ± 07g were distributed in 9 fiberglass tanks (2 m3) randomly after 2 weeks of adaptation with experimental conditions. Feeding was performed three times daily (08:00, 14:00 and 20:00) for 6 weeks for satisfaction. At the end of the period, blood samples were taken and their serum was separated. Serum thyroxine and triiodotyronine were measured by radioimmunoassay in blood samples. Serum thyroxine, triiodotyronine and thyroxine/triiodotyronine level showed no significant change related to dietary OFO. Our findings showed that dietary-oxidized oil had no effect on blood thyroid hormones levels.

Abstract

Gastrointestinal ontogenetic studies constitute one of the basic and important investigations related to the nutrition of aquatic animals. In this investigation, specimens of the western white shrimp (Litopenaeus vannamei) at different developmental stages (from nauplius 1 to postlarvae 120) were assayed for the activities of digestive enzymes. According to the results, at all developmental stages, trypsin, amylase, and lipase enzymes were found to be active. In addition, the peak activities of all enzymes were revealed to occur during the late zoea larval stages (Z3). On the other hand, minimum activities were observed to occur at metamorphosis. During the postlarval developmental stages, amylase and lipase activities increased steadily, whereas the trypsin activity was more or less constant up to the eighteenth week. In conclusion, ontogenetic change in digestive enzyme activity may reflect either a developmentally cued change in enzyme synthesis or a secondary effect of change in the function and relative size of the midgut during its differentiation.

Abstract

Aloe barbadensis is perennial, monocotyledonous, fleshy plant belongs to Aloaceae family. In this study, somoclonal variations of regenerated A. barbadensis plants were investigated. The plantlets of forth subculture transferred to the soil for further study. The genomic DNAs of 40 regenerated plantlets were extracted and genetic variations were studied using SPAR markers including RAPD and ISSR primers. The amounts of Aloe gel also were extracted from regenerated A. vera plants. Average percentage of polymorphism, Shannon index, Nei's genetic diversity and number of effective alleles based on RAPD data were higher than genetic parameters obtained from ISSR data. NJ cluster and STRUCTURE plot based on molecular markers grouped regenerated plants to distinct clusters. AMOVA analysis also showed a significant (P = 0.01) genetic distinction between studied groups. This result also confirmed differentiation of regenerated plants. The amount of Aloe gel in the four groups (based on clustering method) was compared by using analysis of variance (ANOVA). The results showed no significant (P = 0.746) differences between the amount of gel in four group. In total, our findings showed somaclonal variations on genomic level while no significant differences were observed in amount of gel among regenerated Aloe plantlets.

Abstract

The Basolateral Amygdala (BLA) has modulatory effects on working and reference memory. The aim of this study was the evaluation of effects of dopaminergic system in the BLA of rats on working and reference memory behaviors. The number of working and reference errors and time spent in the arms by rats were measured in the radial arm maze according to DSWS protocol. The animals were cannulated in the BLA bilaterally. The microinjection of low dose (0.005µg/rat) and high dose (0.5 µg/rat) of apomorphine have indicated a significant decrease in number of working memory error. But, there was not any change in the numberof reference memory error that showing the improvement of working memory. While the injection of moderate dose apomorphine (0.05 µg/rat) increased these parameters and also enhanced the spent time in working arm that presenting both memories damage. The chlorpromazine injection (2 µg/rat) decreased the number of working and reference memory errors representing the improvement of these memories. Microinjection of chlorpromazine (2 µg/rat) with different doses of apomorphine had no significant change on the both number of errors and the time spent in comparison with control groups. These findings show that BLA dopaminergic system modulates the working and reference memory through that both of (D1/D2) receptors. Also, the effect of this system in BLA is the resultant function of the both receptor families.

Abstract

11, 25 dihydroxy vitamin D3, an active metabolite of vitamin­D3 has been reported to inhibit the growth of number neoplasms such as prostate, breast, colorectal, leukemia and skin cancers. Valproic acid, as a potent histone deacetylase inhibitor, also plays an important role in inhibition of proliferation of tumor cells. However, there are no reports so far on the cooperation between valproic acid and vitamin­ D3 for anti-leukemic effect. The goal of the present research was to evaluate whether low doses of vitamin D3 potentiate the toxicity of valproic acid and whether this toxic action is mediated via apoptotic mechanisms. In this study HL-60 cells were treated either with different concentrations of valproic acid and vitamin­ D3 alone and in combination with each other for 24 hours. Cell survival was determined by MTT assay and then Hoechst staining was used to determine the type of death cell. This present study indicates that vitamin ­D3 potentiates the antitumor effects of valproic acid. Also, the results of staining cells showed that valproic acid and vitamin ­D3 induced apoptosis in HL-60 cells. In total, the new combination of valproic acid and vitamin D3 showed synergistic anti-proliferative effect and induced apoptosis on HL-60 cancer cells.

Abstract

In patients with cerebral ischemia, both during hospitalization and in the community, the prevalence of major depression is evident. Since the depression has a negative impact on recovery, its timely diagnosis and treatment is essential. This study aimed to evaluate the effect of the oral administration of Chevilan extract (Ferulago angulata hydroalcoholic extract; 100, 200 and 400 mg/kg) on brain oxidative stress indices and depression after permanent bilateral common carotid artery occlusion or ischemia/hypoperfusion in male adult rats. A number of 35 rats were divided into a control group, an ischemic group and ischemic groups receiving doses of 100, 200 and 400 mg/kg of Chevilan extracts for 14 days by gavage. To make animal models of permanent cerebral hypo perfusion/ischemia, right common carotid artery was ligatured first and the left one ligatured with an interval of one week. To evaluate depression, immobility in the forced swimming time of each rat was measured and then the rat’s brain tissues were extracted to separate hippocampus and measure malondialdehyde. The results showed that ischemia/hypoperfusion increased brain oxidants such as lipid per oxidation (LPO) and immobility. Also, ischemic rats treated with all three doses of the Chevilan show significant reduction in the concentration of MDA hippocampus and in immobility time at a dose of 100 mg/kg and 200 / 400mg, respectively. Increased swimming time was observed in all three groups of extract recipients. It was found that Chevilan extract with antioxidant effect can reduce the side effects of ischemia such as depression.

Abstract

Strictosidine synthase-like (SSL) is a group of gene families in the Arabidopsis genome, which whose orthologues in other plants are key enzymes in mono-terpenoid indole-alkaloid biosynthesis pathway. The SSL7 is upregulated upon treatments of Arabidopsis plants with signaling molecules such as SA, methyl jasmonate and ethylene. To find the functional role of the gene, a T-DNA-mediated knockout mutant (ssl7) along with the wildt ype were treated with different concentrations of NaCl. The expression level of salt stress genes including P5CS1, NCED3, AAO3 and RD29A at 150 mM NaCl demonstrated that the expression was significantly higher in ssl7 compared with the expression in Col-0. The activities of Catalase (CAT), Ascorbate Peroxidase (APX), Peroxidase (POD) and Superoxide Dismutase (SOD) were measured in different concentrations of NaCl. The results suggested that the enzymes activities were significantly higher in ssl7 compared with wild-type Col-0. In total, the results suggest that SSL7 might have a salicylic acid-dependent negative regulatory role in plant resistance to salt stress.

Abstract

. In this study, morphological variation of three species of Squalius in Iranian basins was studied. For this purpose, 709 specimens were captured from the Caspian Sea, the Urmia Lake, the Namak Lake and the Tigris basin. After anesthetizing in clove oil solution and fixing in 10% neutralized formalin, specimens were transferred to the Isfahan University of Technology Ichthyology Museum (IUT-IM) for further studies. In the laboratory, some 14 meristic characters were counted under a streomicroscope. Also, images were analyzed with ImageJ software and 19 morphometric characteristics were measured. In addition, 13 landmarks were defined and digitized on images taken in tpsDig2 software in order to extract geometric morphometric data. Kolmogorov-Smirnov, Kruskal-Wallis, ANOVA, Duncan test, PCA, CVA and cluster analysis methods were used to analyze the differences among the populations. The results of PCA and CVA meristic and morphometric analyses showed no significant differences among the studied populations. The populations of the species studied were significantly different in 11 meristic and 15 morphometric characteristics. Also, the major differences observed in the results of geometric morphometric analysis were related to the position of the pectoral fin, body and head depth. In general, the studied populations highly overlapped, and we suggest that the populations of genus Squalius should be further studied by molecular methods.

Abstract

Parkinson is a long-term degenerative disorder of the central nervous system that mainly affects the motor system. Pain and emotional disorders due to Parkinson negatively affect the quality of the patient’s life. Biarum carduchrum is an antioxidant plant with some application in traditional medicine. The aim of this study is to evaluate the protective effects of Biarum carduchrum extract on pain and emotional disorders caused by 6-hydroxydopamine injection. Rats were randomly divided into 5 groups of 8 animals. The control group received normal saline. Parkinson's groups were subjected to the injection of 6- OHDA in the right anterior mid-brain (MFB). In third, fourth and fifth groups, rats received Biarum carduchrum extract at doses of 100, 200 and 400 mg/kg via gavage 7 days after induction of Parkinson for 14 days. On day 15th, behavioral tests including forced swimming test and tail flick were performed. Treatment of Parkinsonian rats with Biarum carduchrum extract at doses of 100, 200 and 400 mg dramatically reduced the duration of immobility time in the forced swimming test. Rats treated by Biarum carduchrum extract at doses of 100, 200 and 400 mg showed significantly increased resistance to pain compared with Parkinsonian rats. The results of this study show that the Biarum carduchrum extract improves depression and pain induced by Parkinson, which is probably related to its antioxidant effects.

Abstract

In this study, the morphology and organization of the retina of Caspian kutum and fish response to ambient light as retinomotor reaction was investigated. The Rutilus frisii subsp. kutum is an anadromous fish and important native fish specimen of Caspian Sea. The specimens were obtained from Shahid Ansari Teleost Reproduction and Culture center (Guilan province, Iran). For light and dark adaptation fish were left in dark and light at least 30 minute prior to death. The eye retina were dissected out and processed for histological and SEM studies. The eye retina of Caspian kutum like most vertebrates, was composed of ten layers. Two types of photoreceptor cells including rod and short single cone were identified. Sagittal and transverse retinal sections did not show any particular patterns of cone arrangement. The retinomotor response observed in adapted retina to light and dark conditions revealed that Caspian kutum strongly reacts to changing light conditions. A remarkable pigment index indicated kutum relied mostly on rod rather than cone vision. The movement of pigment granules and slight change of cone myoid showed that the fish could recognize colors.

Abstract

Impaired motor functions were reported in cholestatic animals. This disorder in the function and death of motor neurons is highly dependent on changes in the environment around astrocytes and the blood-brain barrier, which is moderated by the aquaporin 4 protein. For this reason, the effects of cholestasis on motor cortex histology and morphology and aquaporin 4 protein levels were investigated in this study. Samples were stained by hematoxylin-eosin method. Histological changes in cortical brain were investigated. The amount of AQP4 protein in control, sham, and experimental groups were tested by immunohistochemistry. The thickness of motor cortex in cholestatic samples increased in comparison with the control and sham groups. Also, cholestasis caused wrinkle chromatic nuclei. On the other hand, tissue necrosis was detected in cholestatic group compared with sham and control groups. Reduction of cells densities in some cortical layers has been observed, which is probably indicative of cholestasis-induced cell death. AQP4 expression significantly decreased in BDL (p <0.05), but not in other groups (P<0.05). In this study, the pathology of motor cortex, which has also been associated with the decrease of neurons, could be considered the cause of motion abnormalities and AQP4 level reduction in cholestatic rats.

Abstract

Community Level Physiological Profiles (CLPP) is novel method to evaluate microbial activity and diversity in ecosystems. According to the previous findings, poly-β-hydroxybutyrate (PHB) as a bio-control product, increases bacterial diversity in some aquatic animals. In this study, the effects of four experimental diets (control, combination of two PHB degrading bacteria, 2% PHB, bacteria+ 2% PHB) on the anaerobic activity of cultivable bacteria in Siberian sturgeon fingerlings hindgut was investigated. Community level physiological profile (CLPP) was used to analyze anaerobic metabolism by using Biolog™ Ecoplate microplates. The results indicated that anaerobic metabolic potential of the bacterial community was different in the 2% PHB-treated fishes as compared with other groups and PHB improved anaerobic metabolism of bacteria in fingerlings. Also anaerobic metabolism of bacteria was calculated on the different carbon sources (amino acids, carboxylic acids, carbohydrates and polymers) in Ecoplate microplates and the results showed that fish treated with PHB had the highest metabolic activity in mentioned carbon sources. The results of this research revealed that replacing of the diets with 2% PHB increases anaerobic metabolism of culturable bacteria in Siberian sturgeon hindgut while adding PHB degrading bacteria did not change this parameter in fingerlings.

Abstract

Diabetes mellitus is known to be resistant to insulin, to dysfunction of beta cells and to increase liver glucose production. Poor glucose control during hyperglycemia causes damage to the tissues and creates dangerous consequences, such as infertility. Chronic hyperglycemia has harmful effects on the growth of follicles, which is essential for normal female sexual function. Metformin is the most widely prescribed drug in diabetes, but chemical drugs, despite their undeniable benefits, have destructive effects, so alternative strategies for current modern diabetes medications are essential. Herbal medicines are widely used by patients, based on non-medical recommendations and as blood-glucose-lowering agents, including the nettle. Nettle contributes to lower plasma glucose levels by secreting insulin and increasing the proliferation of beta cells in the pancreas. Therefore, the aim of this study was to determine the effect of nettle extract as a supplement of metformin on ovarian tissue of diabetic model. In this experimental study, 30 female Wistar rats were used. Animals were weighed and randomly divided into 5 groups (n=6).  1) control group  2) diabetic group who were diabetic with intravenous injection of alloxan (150 mg / kg)  3) diabetic group + Nettle root extract (150 mg / kg)  4) diabetic + metformin (150mg / kg)  5) Diabetic group + Metformin(150 mg / kg) + Nettle root extract(150 mg / kg). At the end of treatment, the effect of metformin and nettle root extract on ovarian tissue and biochemical factors such as blood glucose and sex hormones were compared and the data obtained were analyzed by SPSS. Hyperglycemia and body weight loss after metformin and nettle root increased for 4 weeks. Simultaneous administration of metformin and extracts of nettle root significantly increased the primordial, primary, secondary, and corpus luteum and reduced the atretic follicles and significantly increased FSH, LH and testosterone levels as compared with metformin alone. The results of this study showed that the root of nettle with its antioxidant compounds and other properties could be a complement to metformin with a corrective effect on hyperglycemia and the improvement of ovarian disorders.

Abstract

Zebra fish is an important species in genetics and considering the proximity of its genome to the human genome, investigating the expression of some of the growth and appetite genes during its larvae development is essential. Genes coding growth and appetite (GH and ghrelin) hormones are involved in the synthesis and release of growth hormone, which can be considered to be economic genes in pisciculture. Given the importance of these genes during the early larvae development stages, this study was performed to assess their activity. Samples were collected at 4, 7, 10, 15, 30 and 45 post-hatching days. Samples were immediately placed in liquid nitrogen (-196 degree centigrade) and then stored in a freezer at -80 degree centigrade until RNA extraction (using RNX-Plus kit). To analyze normal expression of target genes, reference gene β-actin was used by 2-∆∆Ct method. The expression of genes associated with the growth and appetite was significantly different at various stages of the development of zebra fish, as the gene expression of GH on day 4 and ghrelin gene on day 10 after hatching were significantly higher compared with other samples (P<0.05). Overall, the expression of GH and ghrelin genes at the early stages of growth is higher than other fish developmental stages due to their great importance in the survival of larvae at these stages of life.

Abstract

Nowadays, brown macroalgae are considered to be a source of various nutritional and pharmaceutical compounds. In addition, fish roe is very susceptible to spoilage due to its high amount of protein and unsaturated fatty acids. In order to postpone the deterioration of roe-related products, salt and preservatives are utilized. Natural preservatives are recommended because of the adverse effects of chemical additives. Thus, different concentrations of pure alginate (0.1%, 0.2%, 0.3%, 0.4%, 0.5%) were chose and then the 0.2% alginate was selected on the basis of the sensory evaluation of the results. Moreover, the treatments including control (salt-alginate-free), A1 (containing 2.5% salt) A2 (containing 0.2% alginate), A3 (containing 2.5% salt and 0.2% alginate) were used to investigate their preservative effects on fish roe. The quality of rainbow trout roe was assessed within 0, 15, 30, 45 days at refrigerated storage. The treatment A3 showed significant differences in microbial, chemical and sensory results compared with other treatments (p≤0.05). According to the results, salting 2.5% and alginate 0.2% method gave good flavor and increased shelf life to the roe.

Abstract

Sperm morphometry is a part of the semen analysis based on CASA technology and has shown a big role in the prediction of male fertility. This analysis implies the use of stain techniques, although it has been shown that staining can make dramatic changes in the cell morphometry in different species. The aim of the present work was to evaluate the differences in sperm morphometry in Alpaca, introduced by the use of different stain techniques. Single ejaculates from five adult alpacas were used. Samples were recovered by deferent duct deviation surgery technique. Three stain techniques (i.e. Hemacolor, Harri’s Henatoxylin and Diff-Quik) were evaluated. Morphometric analysis was done using ISAS® v1 CASA-Morph system, at 100x bright field objective magnification and a digital video camera with a final resolution of 0.08 µm/pixel. Almost 200 randomly selected cells were automatically analysed per sample and stain technique, considering several sperm-head and midpiece parameters regarding size and shape. Almost all of the parameters showed different levels of difference among the employed techniques. In general, the largest cell heads were the ones stained with Harri’s Hematoxylin and the smallest ones were the ones stained with Diff-Quik. The discriminant parameters used to differentiate between animals better were the head width, area and acrosome percentage. In conclusion, like in other species, alpaca sperm morphometry results were found to be sensitive to the applied stain technique. This implies the necessity of referring clearly the stain technique used in each case to perform comparisons between different works on the same species. None of the employed techniques was superior to the others. In addition, the method used to obtain the samples showed its usefulness and simplicity for repeated samplings.

Abstract

One of the most important factors related to male fertility is sperm chromatin status. Under Assisted Reproductive Techniques (ART), especially Intracytoplasmic Sperm Injection (ICSI), the natural selection of high quality sperm with intact chromatin is not possible. Therefore, embryo quality and pregnancy rate decreased for damaged DNA of sperms involved in the fertilization of eggs. Thus, this study evaluated the frequency of abnormal sperm chromatin condensation and damaged sperm chromatin in men with different infertility factors (e.g. oligozoospermia, asthenozoospermia, teratozoospermia, oligasthenoteratozoospermia, and normozoospermia) and ICSI outcomes were examined. 195 patients were examined and the sperm chromatin status was evaluated using aniline blue and toluidine blue staining. Fertilization, zygote and embryo quality, chemical pregnancy and abortion rates were calculated, too. The results showed that in the oligoasthenoteratozzoospermia samples, the damaged chromatin percentage was higher than that in normozoospermia ones. The fertilization rate, embryo quality, and pregnancy rate significantly decreased in this group. Also, a higher abortion rate was observed in this group (P<0.05). In conclusion, the frequency of damaged chromatin was observed to be different in different male infertility factors, which could influence the ICSI outcomes. Therefore, the evaluation of sperm chromatin status before cycle initiation was found to promote ICSI outcomes.

Abstract

In order to investigate the differences in the shape of the big-head and two morphotypes of Phytophagous fish by geometric morphometric technique, 30 samples of each group, with a mean standard length of 25 ± 3 cm, were prepared. After fish were anesthetized by 1% Clove powder extract and stabilized in formalin buffer 4%, 6-megapixel digital images of the left profile of the specimens were prepared. A total of 17 morphological landmarks were digitized by tpsDig2 software and the tps files were prepared. All non-shaped data were removed from landmark coordinates by generalized procrustes analysis. After the approval of regression between shape space and tangent space, data were analyzed using Principal Component (PCA), Canonical Variation (CVA) and Cluster Analyzes (CA) methods in PAST and TPS series. According to the results, the head length and height, the size of dorsal fin, the length of pre ventral distance, the height of caudal peduncle and the degree of posterior position of the pectoral fin in bighead carp, are some of the distinctive morphological features among species, mainly related to their ecological niche (specially feeding strategies). In the case of silver carp variants, the posterior position of pectoral fin base in silver carp-R that is close to bighead carp, may be due to hybridization between bighead carp and silver carps.

Abstract

Noise pollution is one of the hazardous factors in the environment. One of the major sources of noise pollution is urban transportation, mostly the subway system and transportation in business centers. In this study, the effects of noise pollution on blood serum proteins level of Wistar male rats were investigated. Rats were divided into 3 groups (1 control and 2 experimental, n=6). The control group was kept in standard conditions and the experimental groups were kept in Tehran Grand Bazaar and the vicinity of Sadeghiyeh metro-station (West Rose Street). After eight weeks, the blood serum proteins of each group were analyzed by electrophoresis assay. Then, the blood serum samples were investigated by Ultraviolet-Visible spectroscopy and chemometrics method (MCR-ALS). Assessment of serum proteins indicated that among 5 essential proteins in the blood, the percentage of alpha-globulins and beta-globulins displayed significant difference (p<0.05) in experimental groups. The UV-Visible spectra and chemometrics method showed that there were different species in both experimental groups as compared with the control group. Thus, the noise pollution of the areas studied led to alterations in blood proteins. Consequently, it seems crucial to take control and prevention measures for noise reduction in these areas.

Abstract

The cave barb habitat is located in a Karst formation along the Sezar River. The springs on the walls of the Sezar River valley may provide a means for fish in surface waters to penetrate into the underground waters. These observations propose the probability for a migratory relationship between Garra gymnothorax in the Sezar River and the cave barbs (Garra typhlops and Garra lorestanensis). In addition, a variety of different body shapes including fusiform and slender body forms are observed among the cave fish. This phenotypical variation may be a sign of an unknown genetic diversity or could be attributed to the variable environmental conditions in different parts of the subterranean habitat. To clarify the situation, we used the sequences of mtDNA cytochrome oxidase subunit I and next generation sequencing method. The results showed that the fusiform and slender body shapes of G. typhlops and G. lorestanensis were not different with regard to their mtDNA and genomic compositions. Moreover, the analysis of the genomic their mtDNA and genomic compositions. Moreover, the analysis of the genomic showed that a limited level of gene flow (less than 3%) from G. gymnothorax probably existed in G. thyphlops. The low level of gene flow may be related to the lower fitness and adaptability of the surface dwelling fish to the subterranean life conditions.
