Abstract

Polyploidy induction in German chamomile (Matricaria chamomilla L.) by herbicide trifluralin

Abstract. Chamomile (Matricaria chamomilla L.) is a medicinal herb belonging to Asteraceae family. This study was conducted to investigate the effect of trifluralin on the induction of polyploidy in chamomile. Two independent full factorial experiments on seedlings and 2-leaf stage apical buds were performed.  Following morphological, biochemical and cytogenetic analysis, chromosome numbers of 18 and 36 were detected for diploid and tetraploid types, respectively. Morphological and biochemical examinations revealed that an increase in the number of full chromosome set results in a reduction in stomata number per unit area as well as an increase in stomata size, chloroplast number, and chlorophyll content. Induction of ploidy level increments also reduced plant height and increased the number of lateral branches, leaf size, and diameters of stems, flowers and receptacles. It is concluded that a concentration of 22.5 µM trifluralin in both methods is optimum for the production of tetraploid chamomile with the highest rate of polyploidy induction and the lowest percentage of abnormality.

Abstract

In this paper, the new taxa Lepidium ferganense (Brassicaceae) was recorded for the flora of Iran for the first time. This taxon belongs to Irano-Turanian region. It was compared with the closest taxa in the present study. In addition, a morphological description, several Illustrations, and a distribution map were presented.

Abstract

Stipa richteriana and Galium songaricum are newly recorded species for the flora of Iran, collected from Binalood and Hezar-Masjed Mountains in Razavi Khorassan Province. The geographical distribution of both species is mainly confined to the Middle Asia. Morphological characters of two newly recorded species are compared with their close relatives. Notes on taxonomy, ecology, phytogeography, and conservation status of both species are provided.

Abstract

Scorzonera incisa, known as an endemic plant to Turkey, has been collected from West Azarbayejan Province, NW. Iran. The species is reported for the first time from the country; it is very similar to Scorzonera calyculata, which is distributed in the western half of Iran and east of Iraq. However, with regard to the particular shape of the achenes of Scorzonera incisa, this species could be separated from Scorzonera incisa. In this paper, the description, the image of the herbarium sample and the distribution map of this species are presented.

Abstract

The distribution patterns of taxa belonging to the section Hypoglottidei DC. were investigated using DIVA-GIS software. Based on Zohary classification, five global patterns, with the exception of widely distributed species, include 1. Centro-Asiatic province, 2. Irano-Anatolian province, 3. Pontic province, 4. East Mediterranean subregion and 5. West Mediterranean subregion, were identified. In addition, the Iranian species of this section were classified in three patterns: 1. Central Zagros 2. Khorasan-Kopet Dagh 3. Central and Eastern Alborz. The maps and distribution patterns of the species were also presented.

Abstract

Nutlets of 20 taxa of the tribe Marrubieae were examined by scanning electron microscopy (SEM) and detailed descriptions of nutlet micromorphological features for all examined taxa are provided. The nutlets exhibited variation in size, shape, color and surface sculpturing. The nutlets shape of most species studied is ovate, but rounded, broad ovate, elliptic, lanceolate, triangular and oblong can also be found in a few species. Six basic types of the sculpturing pattern of nutlet surface can be distinguished: reticulate, foveolate, scalariform, ruminate, pusticulate and colliculate-granulate. The most common type of nutlet sculpturing among the studied species is reticulate, but the variation in size and shape of their composing cells provided useful diagnostic characters. Our investigation revealed that the type of sculpturing was more useful in taxon delimitation among Marrubium species and allies at the species rank.

Abstract

Achene morphology of 49 taxa of the genus Centaurea L. was studied in terms of 19 different characteristics. On the basis of the variation in these features, some sections, such as sect. Cyanus with hairy hilum, were separated. Despite various differences, C. leuzeoides and C. gilanica were categorized in the section Psephelloideae, a section with lots of character variations within its species. Also, C. albonitens has different characteristics in comparison with other taxa within section Phaeopappus, for example features such as pappus color and the ratio of pappus length to that of achene separated the species from other members of the section. The last but not the least, highly different characteristics were observed in the achene morphology of C. phlomoides as compared with other species in section Cynaroides. Differences such as short pappus and hilum length were found to separate C. phlomoides from other members of sect. Cynaroides. In conclusion, on the basis of the findings of this research, most achene morphological characteristics can be appropriately used as key features for the differentiation of sections in the genus Centaurea. An identification key based on the features of the achene, images related to the achene of the studied species were also presented

Abstract

Antioxidant enzymes play an important role in plant defense against pathogenic agents. Following the identification of the pathogen, plants produce active oxygen species (ROS) as one of their first defense responses. To maintain the balance of ROS levels and prevent their harmful effects, plants produce antioxidant peroxidase (POX), catalase (CAT), ascorbate peroxidase (APX) and superoxide dismutase (SOD) enzymes. In the present study, the resistance of bean plants cultivars, namely Sadri, Paak, Darakhshan and Dorsa, to Xanthomonas axonopodis pv. phaseoli (Xap) were studied in greenhouse conditions. The catalase, peroxidase and ascorbate peroxidase enzyme activities were studied in healthy and Xap-infected bean cultivars Sadri and Derakhshan at 0, 24, 48, 72 hours and 20 days post inoculation by a completely randomized design with 5 treatments and 4 replications. The result showed that disease symptoms appeared in all tested cultivars. Derakhshan and Sadri cultivars, with 58.33 and 80.56 percentages of infected plants 20 days after inoculation, showed the least and highest infection rates, respectively. The highest catalase and peroxidase activities were recorded 24 and 48h post inoculation. These records reduced 48 and 72 hours post inoculation, respectively. The activities of these two enzymes in the susceptible cultivar were less than those in the semi-resistant one. The chlorophyll a and chlorophyll b contents of Xap-infected plants reduced significantly. The total chlorophyll content of uninfected Sadri and Darakhshan cultivars were 2.93 and 3.23 µg/g, respectively, which reduced to 1.96 and 2.14 µg/g of leaf tissue in infected plants, respectively. Based on these results, it is suggested that the Derakhshan cultivar should be planted in disease-susceptible regions as the semi-resistant cultivar.

Abstract


To investigate the effect of herbicide Trifluralin and the role of magnetized water on enzymatic activities and content of proline and growth substances including gibberellin (GA3) and cytokinin (6- benzyl amino purine), an experiment was conducted using squash (Cucurbita pepo L. var. Shiraz Hybrid F1) seedlings. Four levels of trifluralin (0, 5, 15 and 25 ppm) and irrigation by distilled and magnetized water were applied in a completely randomized design in 3 replicates at Urmia University in 2016. Different levels of trifluralin were added to pots 2 days after planting. Seedlings were treated by modified Hoagland solution with diurnal temperature 30:18 °C, relative humidity of 70-80 percent and light period of 16:8 hr, during a 5-week growing period in growth chamber. The effect of magnetized water and trifluralin on ornithine amino-transferase (OAT), proline dehydrogenase (PDH) and content of proline in both shoots and roots as well as GA3 and cytokinin was significant (p>0.01) according to ANOVA. The content of proline and OAT in roots and shoots as well as GA3 and cytokinin increased significantly, although the PDH of roots and shoots decreased under the influence of magnetized water. The content of proline and OAT increased and GA3, cytokinin and the activity of PDH declined by the increase of the levels of trifluralin. Overall, it can be concluded that the growth and biochemical indices of squash plants were improved by magnetized water under toxicity of herbicide trifluralin.

Abstract

The cement production activities leads to the release of different pollutants into the environment. This research was conducted to study the effects of dust particles released by Behbahan cement factory on the absorption of some nutrient elements and biochemical parameters in dominant herbaceous species grown in this area, including Bromus tectorum, Malva neglecta and Sinapis arvensis. Plant samples were transferred to the laboratory. Then, the concentrations of some nutrient elements, such as Fe, Cu, Zn, K, Mn, P and N, were assayed in plant and soil samples. Moreover, some biochemical parameters, such as photosynthetic pigments, total protein, soluble carbohydrates, proline, relative water content, leaf pH total ascorbate and air pollution tolerance indexes (APTI), were evaluated. Based on these results, it is supposed that three plant species are tolerant to air pollution with an APTI higher than 16. In addition, it seems that the dust released by the cement factory has not led to a toxic accumulation of the elements in the studied plants. The deficiency of Mn and P was determined for three plant species. On the basis of biochemical analysis, it is supposed that these plant species possibly use different strategies to tolerate the pollutants in this area.

Abstract

The purpose of this research was to identify the structural adaptations in three populations of Convolvulus persicus L. on coastal areas in Mazandaran Province, whose populations of which are severely reducing due to the intense environmental stresses of the coastal zone. In general, annular collenchyma, isobilateral mesophyll, amphiphloic siphonostele, abundant presence of laticiferous tubes, numerous druse crystals and the presence of periderm in rhizome can be introduced as the most important morpho-anatomical strategies utilized against harsh environmental conditions. Sari population was foud to have the most various structural toleration mechanisms in comparison with Babolsar and Nour populations. Few differences were observed in anatomical characteristics in the three populations of Convolvulus persicus as the evidence for high intra-specific phenotypic variability, leading to local adaptation and increase of the tolerability of plants against environmental changes.

Abstract

Aloe vera L. is one of the most valuable plants in the pharmaceutical, cosmetic, sanitary and food industries. In vitro culture is used for commercial production and due to the abundant application of this plant, extensive research has been performed on the in vitro culture of Aloe vera. For this purpose, the present study was conducted at two stages. At the first stage, the best method of sterilization of explants derived from Aloe vera offshoots was investigated. At the second stage, the effect of the type of explants, the light condition (dark and bright) and the effect of BAP (Benzyl Amino Purine) and NAA (α-Naphthalene acetic acid)) growth regulators on regeneration and the amount of phenolic compounds were studied. A factorial experiment was executed on the basis of a completely randomized design with three replications. The best sterilization protocol was 0.1% mercuric chloride (for 2 minutes), 70% ethanol (for 30 seconds) and 15% sodium hypochlorite (for 5 minutes). The little white explant derived from the base of leaves, with the lowest percentage of phenol and the highest survival rate (67.5%) in darkness, was found to be the best candidate. MS medium supplemented with 0.75 mg / L BAP and 0.25 mg / L NAA resulted in the highest stem number (2.5) and stem length (42.107 mm), establishment percentage (73%), leaf number (6.33), leaf diameter (4.8 mm), chlorophyll b (9.216 mg/g) and carotenoids (4.81 mg/g). The highest content of chlorophyll a (56.07 mg/g) and total chlorophyll (61.35 mg/g) were found in samples treated with hormonal medium, supplemented with 1.5 mg / L of BAP with 0.5 mg / L of NAA. The maximum number (3) and average length (33.3 mm) of roots were observed in samples treated with the hormone-free medium.

Abstract

The phylogenetic relationships among 39 species belonging to 12 sections of Astragalus from Iran were studied on the basis of 29 morphological characters. The cladistics analysis of the morphological data was performed using PAUP* 4.0b10 program. The obtained data were compared with the molecular systematics data obtained from nuclear DNA ITS. In contrast with previous molecular systematic studies, the morphological data placed the sect. Caraganella as paraphyletic clade to the sects. Cenantrum, Nuculiella, and Eremophysa. Our results were similar to those obtained by previous molecular systematic studies which specified sects. Astragalus, Alopecuroidei and Laxiflori as paraphyletic clades. In accordance with data presented in previous molecular systematic studies, the species of the sect. Caprini showed close relationship with those in sects. Pelta and Pendulina. The present analysis, consistent with molecular systematic studies, rejected the monophyly of most sections studied. Therefore, although the circumscription of those sections needs to be carried out on the basis of the results of widely accepted molecular and morphological phylogenetic studies, future studies are needed to clarify and resolve some remaining ambiguities.

Abstract

Charcoal rot caused by Macrophomina phaseolina is one of the most important soil borne diseases with a broad host range including bean, which annually brings a significant damage to this plant. Biological control of charcoal rot is very important because its chemical control harms the environment, microflora and soil fertility. Chemical control of charcoal rot is also difficult and sometimes ineffective. Fluorescent Pseudomonads are able to increase plant growth and inhibit the development of plant pathogens by producing and secreting antibiotics, enzymes, siderophores, and plant hormones. In this study, infected bean plants by M. phaseolina were collected from infected bean fields of Khorramabad (Lorestan Province, Iran) in the summer of 2015. Virulence of fungal isolates was evaluated in a greenhouse and one isolate with the highest pathogenicity was chosen for further experiments. The biocontrol potential of eight Pseudomonas fluorescens strains, whose biocontrol abilities were proved in previous studies, was examined against M. phaseolina in vitro. The growth inhibition of M. phaseolina was examined by dual culture test and the antifungal activity of bacterial volatile and nonvolatile metabolites. P. fluorescens UTPf125, which showed the highest inhibitory effect on the mycelial growth, was selected for greenhouse tests. UTPf125 strain led to a significant reduction (%50) of disease severity and increased fresh and dry weight significantly. Phenol compounds were evaluated 1, 3, 5, 7 and 9 days after inoculation by pathogen. The results showed that the highest value of total phenol content was obtained on the third and fifth days after inoculation, decreasing on the seventh and ninth days.

Abstract

The present study aimed to increase the seed germination and the initial performance of pistachio seeds by investigating the effects of the magnetic field and osmopriming treatments simultaneously. 10 treatment levels of 10, 20 and 30 mT for 5, 15, 25 minutes and 1 control treatment (without exposure to a magnetic field) were performed in three replicates and the best level of magnetic surface was selected. The results showed that seeds subjected to the magnetic field showed an increase in plumule length, radicle length, total length of seedlings, fresh and dry weight of plumule as well as radicle and germination speed compared with the control treatment. However, there was no significant difference in the germination percentage and uniformity of seeds. Magnetized seeds, with the superior selective treatment (10 mT for 15 minutes), were primed with solutions of potassium nitrate, humic acid and salicylic acid at levels of 10, 25 and 40 mM and a control level (the best level of magnetism) in three replicates. The best result was obtained from humic acid (25 mM). With regard to the low cost of the purchase or construction of magnetic devices and their positive effects, their application could be recommended (10 mT for 15 minutes) combined with the humic acid (25 mM)) so that the quality of pistachio seedlings production would be increased.

Abstract

Pterocephalus (Caprifoliaceae) consists of 30 species worldwide, mainly distributed in the temperate regions of the northern hemisphere, of which 14 species were reported from Iran, including five endemic ones. In this study, 35 leaf-related traits and 30 peduncle-related traits were investigated by light microscopy of 15 samples from 11 Iranian species of Ptreocephalus. Our results showed that there were two types of midrib, six types of leaf indumentum and five types of peduncle indumentum. Crystalized calcium oxalate was observed in the leaves and peduncles of most species except in P. plumosus and P. brevis. The profiles of the peduncles were hexagonal, circular or elliptical. The vascular cylinder shape varied from circular to sinuate form. P. wendelboi, with a very different morphology as compared with other species, also exhibited different anatomical characters. The hexagonal sections of peduncle in P. ghahremanii and P. kurdicus and two samples of P. szovitsii were found to be very similar. However, the mentioned characters were found to be less informative or charachterisation of other species. In conclusion, the results showed that the anatomical characters of the leaves and peduncles might have some taxonomic importance in the identification of some species of the genus Pterocephalus.

Abstract

One of the main strategies to improve plant tolerance is the expression of stress-induced genes, which play a significant role in the ionic balance of plants. SOS3 is one of the important components of SOS-regulated ionic homeostasis pathway. Therefore, the expression of this gene could be an important step towards producing salt-resistant plants. In this work, we have transformed tomato (Solanum lycopersicum) by Agrobacterium (GV3101 and LBA4404) containing plasmids with SOS3 genes. The maximum regeneration rate was determined in cotyledons of CH genotype. The simultaneous use of cotyledons and hypocotyls in the culture medium had the best outcome. In addition, the best time was found to be one day after inoculation. Also, the best transgenic variety was detected for Agrobacterium GV3101, which can be attributed to the interaction between the genus Agrobacterium and the tomato variety. Transgenic plants were transferred to a culture medium containing sequestrene, which caused the acceleration of the seedling growth in particular. The presence of the SOS3 in the transgenic plants was verified by PCR and RT-PCR methods.

Abstract

Floristic study is one of the most efficient methods of gaining knowledge about biological capacities, as well as management and conservation of plant genetic resources in ecological systems. Zanjan county town is located in a mountainous region with an area of 6763 km2 and the average altitude of 1663 m.a.s.l. Its favorable vegetation diversity is the result of suitable ecological conditions, diverse habitats and appropriate water resources in the region. The central district of Zanjan county town, with an area of 2600.2 km2, was chosen as a representative to show tree and woody shrub species diversity. In order to identify tree and shrub species, life forms and chorology of plants, samples were collected by the conventional method of floristic studies and were, then, identified using taxonomic references. In total, there were 122 tree and woody shrub species, belonging to 76 genera and 39 families, in the studied area. The richest families included Rosaceae (29 species) and Oleaceae (9 species), which comprised 31.15% of the arboreal and woody shrub species. Mesophanerophytes (37.7%) and microphanerophytes ‌(32.79%) ‌were the dominant life forms in the area studied as determined by the Raunkiaer method. The Irano-Turanian vegetative elements were the most important elements in this area based on Zohary approach. Prunus lycioides was found to be endemic to Iran.

Abstract

Fire affects vegetation and changes plant succession. In this paper, the vegetation of Sirachal Research Station, which burned in the summer of 2014, was studied and compared with the unburnt areas, based on biodiversity indices. The research was implemented as a factorial experiment in a completely randomized design. First, on the basis of physiognomy, the area was divided into three parts: shrubland, shrubland-rangeland, and rangeland. Each part was, then, divided into two sections, including a burnt area and an adjacent unburnt area (control area). In each area (burnt and unburnt), three sampling units were randomly assigned using PNP method, and vegetation measurements were subsequently performed. Based on data analysis, a total of 141 taxa were identified, belonging to 28 families and 95 genera, including one species of Gymnosperm, 19 monocotyledons and 121 dicotyledons in two burnt and unburnt areas of Sirachal Station. Based on the statistical analysis, using Past3 and SPSS softwares, there was no significant difference in the number of taxa between the vegetation of the burnt and unburnt areas. Of the indicators analyzed in Past3, the number of individual, the index of Menhinick's richness and the Shannon diversity index were significantly different in the treatments sampled from the burnt and unburnt areas. According to the Duncan test average, the number of individuals in the unburnt area, with a canopy cover below 25%, was greater than that in the burnt area, with the same canopy cover, however, no significant difference was observed between the individuals in the burnt area and unburnt area, with the same canopy cover of more than 25%. Also, the richness of the burnt area with a canopy of less than 5% is significantly higher than that in the unburnt area with the same canopy cover.

Abstract

The present study was aimed to investigate foliar anatomy and flower, fruit and seed morphology of three subspecies of Agrimonia eupatoria (i.e. A. eupatoria subsp. eupatoria, A. eupatoria subsp. grandis and A. eupatoria subsp. asiatica) and the species of Aremonia agrimonioides, belonging to the subtribe Agrimoniiae (Rosaceae) in Iran. Firstly, plant specimens were collected and then identified on the basis of relevant references. For anatomical analysis, the cross sections of basal leaves of each taxon were prepared, studied by light microscopy and then photographed by means of a digital camera. For morphological analysis, the flower, achene and seed characters were carefully examined using digital microscopy. To determine the relationships among the taxa, 57 characters were included in the Principal Component Analysis (PCA). The results supported the classification proposed by Flora Iranica and showed the importance of leaf anatomical, flower, achene and seed morphological characters in separating inter-generic, intraspecific and interspecific relationships among the representatives of subtribe Agrimoniinea in Iran.
