Abstract

Tea was planted in Lahijan by Kashefalsataneh in 1930. The main concern about important commercial plants such as tea is the formation of ice crystals in low temperatures. This can damage the live cells leading to lowering the quality of the plant and eventually its death. Formation of reactive oxygen species (ROS) and oxidative stress is the result of various environmental stresses leading to freezing. Investigating the variations in any of these factors could help to understand the mechanism of freeze resistance in ever-green plants. The aim of the present research was to investigate lipid peroxidation, the presence of antifreeze protein and variations in the activity of some antioxidant enzymes, including superoxide dismutase (SOD), ascorbate peroxidize (APX) and catalyse (CAT) in tea leaves subjected to 20, 0, -2, -5 and -8°C in tea leaves from the north of Iran. The results showed formation of an antifreeze protein with MW of about 20 KD in response to cold stress. It was also found that the activity of SOD, APX and CAT increased in tea leaves due to cold stress. The activity of SOD increased down to -8°C. APX and CAT increased their activity down to -5°C. On the other hand, the lipid per oxidation factor, MDA, was also elevated in response to the cold stress.

Abstract

 Specimens collected from the forest areas of Mazandaran Province (Iran), Kojour, between Lashkenareh

and Gandis-Kala village showed significant characteristics. At first glance, these specimens looked similar to A. szowitsianum, but with no indumentum on fruit. Further studies showed that those specimens belong to a new species, Alyssum mazandaranicum Mirzadeh & Assadi sp. nov., which was confined to the ecotone area between Euro-Siberian and Irano -Turanian regions. More populations of this new species were also found in other localities with similar ecological circumstances. Here, the new species is described, illustrated and compared with the closest taxon.

Abstract

Garra rufa is one of the 73 Garra species of the family Cyprinidae. Scales are used more commonly than any other structure in population dynamic studies, because of their transparency, ease of sampling, and the minimal in-juries caused to the fish during their removal. To discriminate fish from different rivers including Bashar, Behbahan, Gerdab, Kheirabad, Kooshk-e-Bahram, Mazoo, Palangan, Sendegan, Shoor-Abad, and Sirvan Rivers using geometric morphometric analyses of the scale shape, landmark based geometric morphometrics method was used. Seven landmark points were depicted on each scale using the software TpsDig2. After procrustes analyses, landmark data were exposed to principle component analysis (PCA), CVA, and cluster analyses using the past software. The results showed that th-ere were significant scale shape differences among some populations, but most of the samples showed overlapping sh-ape distribution and could not be separated robustly. Among the samples which were analyzed, only the Sendegan and Gerdab river samples did not show significant difference in scale shape compared to all other samples.

Abstract

Floral morphology of 24 species of the genus Alchemilla in Iran was studied by means of light and digital mi-croscopes. Flower samples were collected from different species and their morphological traits, including hypanthium, calyx, epicalyx, pedicels, ovaries and stamens, were measured. The most useful features for the separation of species were turned out to be the calyx shape and length, trichomes position on calyx and pedicle. Principal Component Anal-ysis (PCA) and Cluster Analysis (CA) were used to determine the potential contribution of floral morphological char-acters in the expression of species relationships. A total of 17 qualitative and quantitative characters were utilized in both analyses. The results of UPGMA and PCA methods were found to be consistent and in agreement with current classifications.

Abstract

Thirty eight ecotypes of watermelon were collected from different parts of Iran. After the preparation of the field, these eotypes were cultivated in a completely randomized block design with three replications. In order to invest-igate genetic diversity, genomic DNA samples were extracted from leaves and Polymerase chain reactions were optimized using 14 SRAP primer pairs. One hundred thirty six polymorphic bands were detected, of which the EM10-Me4 was the most abundant primer pair with 19 bands and EM16-Me4 and EM16-Me14 were the least primer pairs with 7 bands. PIC index varied from 0.20 to 0.32 and genetic diversity was 0.17 to 0.28 on the basis of Nei index. Fisher's Linear Detection Analysis showed that the UPGMA method and the grouping accuracy of about 90% are more appropriate than other cluster analysis methods. Cluster analysis, using Jakard method, was performed and the ecotypes studied were classified into five distinct groups. Based on the PCA, the first and second components included 92.5% of the variation, which represents the proper distribution of the markers on the whole genome.

Abstract

The genus Zoegea L. belongs to Asteraceae family and has about 10 species in the world. This genus is considered to be an Irano-Turanian and Mediterranean element and is distributed in south-western and central Asia and in the central, southern, north-western and south-western parts of Iran as well. The subspecies classification of the genus is not consensus and various classifications could be found in different taxonomy resources. In this study various specimens from different regions of Iran were studied. In addition, anatomical and palynological characters were used to perform a cluster analysis in order to determine species groups. In the end, our results confirmed that Z. baldschuanica and Z. glabricaulis were distinct species.

Abstract

The genus Ganoderma belongs to Polyporales, well-known for its medicinal properties and as a plant pathogen. The aim of this study was to identify the genetic variation between populations of two Ganoderma lucidum Karst. complex isolates using RAPD. For this purpose, four populations of G. resinaceum Boud. and eight populations of G. lucidum collected and purified during 2011-2012, were studied for the first time in Iran. Ten random primers were used for RAPD reaction. The result for RAPD-PCR studies of G. lucidum complex showed a wide range of diversity, as scores of 61.48 and 40.16 were estimated for the inter-specific diversity of G. lucidum and G. resinaceum, respectively. Cluster analysis confirmed the separation of two species populations in different clusters. In the analysis of genetic structure, two probable groups (K = 2) were identified in the studied germplasm which was also confirmed by the results of bar plot.

Abstract

Sperm morphometry is a part of the semen analysis based on CASA technology and has shown a big role in the prediction of male fertility. This analysis implies the use of stain techniques, although it has been shown that staining can make dramatic changes in the cell morphometry in different species. The aim of the present work was to evaluate the differences in sperm morphometry in Alpaca, introduced by the use of different stain techniques. Single ejaculates from five adult alpacas were used. Samples were recovered by deferent duct deviation surgery technique. Three stain techniques (i.e. Hemacolor, Harri’s Henatoxylin and Diff-Quik) were evaluated. Morphometric analysis was done using ISAS® v1 CASA-Morph system, at 100x bright field objective magnification and a digital video camera with a final resolution of 0.08 µm/pixel. Almost 200 randomly selected cells were automatically analysed per sample and stain technique, considering several sperm-head and midpiece parameters regarding size and shape. Almost all of the parameters showed different levels of difference among the employed techniques. In general, the largest cell heads were the ones stained with Harri’s Hematoxylin and the smallest ones were the ones stained with Diff-Quik. The discriminant parameters used to differentiate between animals better were the head width, area and acrosome percentage. In conclusion, like in other species, alpaca sperm morphometry results were found to be sensitive to the applied stain technique. This implies the necessity of referring clearly the stain technique used in each case to perform comparisons between different works on the same species. None of the employed techniques was superior to the others. In addition, the method used to obtain the samples showed its usefulness and simplicity for repeated samplings.

Abstract

The phylogenetic relationships among 39 species belonging to 12 sections of Astragalus from Iran were studied on the basis of 29 morphological characters. The cladistics analysis of the morphological data was performed using PAUP* 4.0b10 program. The obtained data were compared with the molecular systematics data obtained from nuclear DNA ITS. In contrast with previous molecular systematic studies, the morphological data placed the sect. Caraganella as paraphyletic clade to the sects. Cenantrum, Nuculiella, and Eremophysa. Our results were similar to those obtained by previous molecular systematic studies which specified sects. Astragalus, Alopecuroidei and Laxiflori as paraphyletic clades. In accordance with data presented in previous molecular systematic studies, the species of the sect. Caprini showed close relationship with those in sects. Pelta and Pendulina. The present analysis, consistent with molecular systematic studies, rejected the monophyly of most sections studied. Therefore, although the circumscription of those sections needs to be carried out on the basis of the results of widely accepted molecular and morphological phylogenetic studies, future studies are needed to clarify and resolve some remaining ambiguities.

Abstract

Noise pollution is one of the hazardous factors in the environment. One of the major sources of noise pollution is urban transportation, mostly the subway system and transportation in business centers. In this study, the effects of noise pollution on blood serum proteins level of Wistar male rats were investigated. Rats were divided into 3 groups (1 control and 2 experimental, n=6). The control group was kept in standard conditions and the experimental groups were kept in Tehran Grand Bazaar and the vicinity of Sadeghiyeh metro-station (West Rose Street). After eight weeks, the blood serum proteins of each group were analyzed by electrophoresis assay. Then, the blood serum samples were investigated by Ultraviolet-Visible spectroscopy and chemometrics method (MCR-ALS). Assessment of serum proteins indicated that among 5 essential proteins in the blood, the percentage of alpha-globulins and beta-globulins displayed significant difference (p<0.05) in experimental groups. The UV-Visible spectra and chemometrics method showed that there were different species in both experimental groups as compared with the control group. Thus, the noise pollution of the areas studied led to alterations in blood proteins. Consequently, it seems crucial to take control and prevention measures for noise reduction in these areas.

Abstract

This study was conducted to understand the morphological variation of four populations of Schizothorax pelzami from Iranian inland waters. For this purpose, a total of 81 specimens were collected from the Bidvaz, Cheshmeh-Ali, Kalat and Aal rivers. After anesthesia, all specimens were fixed into buffered formalin and transferred to the laboratory. Then, the left sides of all individuals were photographed. To extract the body shape data, 14 landmarks-points were defined and digitized on 2D image using TpsDig2 software. To eliminate size, direction and position from data, the Generalized Procrustes Analysis (GPA) was performed. Then, PCA, MANOVA/CVA and cluster analysis were used for further investigation. The results showed that there was no significant difference but some overlapping among the studied populations on the basis of PCA analysis was observed. The CVA graph separated the studied populations from on another. The main differences in body shape of the studied populations were related to the position of the snout, the depth of head and body, and the length of caudal peduncle. The observed changes can be considered to be an adaptation to their habitat, which has been influenced by different environmental parameters.

Abstract

In this study, the phylogeny and morphological variation of 6 species of the genus Cousinia sect. Stenocephalae, i.e., C. hypopolia, C. decipiens, C. esfandiarii, C. renominata, C. stahliana and C. daryoushiana, distributed in NE regions of Iran, were investigated. ITS, ETS and rpl32-trnL (UAG) sequences for 12 species (17 specimens) were generated. Phylogenetic interspecies relationships were constructed by Bayesian method and likelihood analysis. The specimens were also investigated in terms of morphological characters. Despite very similar morphological characters and geographical distributions, the separation of these species was confirmed by the obtained results of the molecular data (combined data of plastid and nuclear sequences). On the basis of the present study, the above-mentioned species were almost separated from each other by morphological characters and molecular data. A diagnostic key and the description of these species were also presented. These results did not conform with the previous palynological and anatomical studies of the species.

Abstract

Breast cancer is the fourth common cancer worldwide and occurs when breast cells begin to uncontrolled division and tumor formation. Angiogenesis is one of the essential factors in cell growth and maintenance of homeostasis in the natural and pathological conditions, while VEGFs are the most critical factors in angiogenesis. MiR-210 plays an important role in the angiogenesis via association with VEGF. Here, the miR-210 expression changes in response to a VEGFB antagonist peptide, called VEGB1, was studied in female BALB/c mice bearing 4T1 cell line induced breast tumor. The treated group received 1mg.kg-1 and 10mg.kg-1 of the peptide and the control group received PBS intraperitoneally during two weeks. Both of the animal groups underwent a resection of breast tissue 14 days after treatment and miR-210 expression level was investigated. Statistical analysis by On-way ANOVA showed that the expression level of miR-210 gene had significant differences among the groups treated with various doses of VEGB1. Also, the gene expression was significantly different between peptide-treated groups and control samples (p<0.05). MiR-210 expression level had 42% reduction in mice treated with 1mg.kg-1 of VEGB1, while 90% was seen in mice treated with 10mg.kg-1 of VEGB1 showing the inhibitory function of VEGB1 antagonist peptide at different doses.

Abstract: Due to the presence of taxonomic ambiguity regarding Petroleuciscus ulanus and Alburnus atropatenae and the tendency to treat them as synonyms of each other, this study was conducted to validate these two species by comparing their morphometric and meristic characters in the Mahabad-Chai River, Urmia Lake basin. A total of 64 specimens of P. ulanus and 68 specimens of A. atropatenae were sampled and were then investigated for 17 morphometric and 7 meristic characters. Measurements were standardized to eliminate the effects of size due to allometric growth pattern, then their normality and abnormality were examined. Standardized data were analyzed using t-test, Mann-Whitney, principal component analysis (PCA) and discriminative function analysis based on P-value of Hotelling's t-test (DFA/Hotelling's T-test) methods. The results showed that the two species were significantly different in 11 morphometric and 2 meristic traits (P<0.05). DFA analysis also showed significance differences between the two studied species. Our results refute the synonymy hypothesis of Petroleuciscus ulanus and Alburnus atropatenae on the basis of morphometric and meristic traits. Therefore, based on the results of this study, the taxonomic identities of these two species are valid.

Abstract

Gambusia holbrooki Girard, 1859 is considered as one of the non-native and invasive species in the inland waters of Iran. This species occurs in most inland basins and aquatic bodies of Iran and has even been found in brackish waters. Extensive adaptations of this species to its different habitats caused numerous changes in its biological characteristics and morphological traits. The present study was performed to investigate the sexual dimorphism of G. holbrooki in the Gamasiab River, Kermanshah Province, Iran. 59 fish specimens were collected from the river in September 2017. After taxonomic confirmation and sex determination, 29 morphological traits were measured in fish samples using a caliper and their meristic traits were recorded. In total, 37 characters (including 8 meristic traits and 29 morphometric traits) were examined. After obtaining morphological data, Principal Component Analysis (PCA) and Discriminant Function Analysis (DFA) were utilized to compare morphological traits between male and female individuals. The results showed that the 59 specimens were comprised of 30 malea and 29 femalea. On the basis of the results, all morphological traits, except the caudal peduncle length, dorsal fin base length and a post-anal length, showed significant sexual dimorphism (p <0.05). The results of DFA and PCA analysis showed that males and females were completely different from each other in terms of morphometric traits. These findings indicate that there is a sexual dimorphism in G. holbrooki of the Gamasiab River. The sexual dimorphism observed in this fish species is due to the sexual selection and increase of reproductive success.

Abstract

This study investigated the potential of aquatic bacteria for their ability as a biocatalyst to synthesized Fe2O3 nanoparticles using iron precursor, FeCl3. A total of 25 aquatic bacterial strains were isolated in trypticase soy agar plus 10 mM FeCl3 with selective enrichment technique. Among the bacterial strains evaluated, NV06 was the only strain able to synthesize Fe2O3 nanoparticles extracellularly. The strain NV06 was identified as Alcaligenes sp., on the basis of phenotypic and molecular characteristics. Extracellular synthesis of Fe2O3 nanoparticles by this strain was investigated under the optimal conditions. The biosynthesized Fe2O3 nanoparticles were characterized using UV–visible spectrophotometry (UV-Vis), Scanning electron microscopy (SEM), Energy-dispersive X-ray spectroscopy (EDX), and Fourier transform infrared (FTIR) spectroscopy. The results showed that cell-free extract (CFE) of the bacterium strain can produce the rod-shaped Fe2O3 nanoparticles with mean edge lengths of 80.2 nm and mean diameters of 25.5 nm, after being exposed to FeCl3 solution (10 mM), at an optimum pH of 6 and an optimum temperature of 28 °C, after 96 hours of incubation at 150 rpm. This is the first report on the extracellular biosynthesis of Fe2O3 nanoparticles using the genus of Alcaligenes under the CFE strategy. It could be speculated that the results of the study can hopefully introduce the inherent capabilities of aquatic microbes as safe, simple, and effective biocatalysts in the production of Fe2O3 nanoparticles.

Abstract

The seed micromorphological characters of 15 taxa of Iranian Pyrus comprising 22 individuals were examined by Scanning Electron Microscopy (SEM) and Digital Microscopy (DM) for the first time in this survey. For the assessment of probable ecological effects, several individuals of some widespread species were studied. After collecting several samples of each wild taxa of Pyrus from different localities in a ten-year period, the specimens were identified by means of the examination of type specimens and different Floras. After the examination of the seeds surface, qualitative (e.g., shape and ornamentation of seeds including areolate, foveate, reticulate, colliculate, striate and undulate types) and quantitative characters (e.g., size of seeds, size of surface cells and thickness of cell walls) were assessed. Statistical analyses were performed on the data using SPSS software. According to these observations, seed morphological characters showed to have a considerable variation even among the different individuals of the same species, verifying complexity of the genus as revealed by the different approaches which were previously reported. However, some descriptive characters of seeds such as shape, size, surface ornamentation type, cell size and cell wall thickness are presented for the first time for the genus.

Abstract

Stachys sect. Satureoides includes four species characterized by being annual herbs having oblanceolate leaves, urceolate calyx in fruit and dense hairs at mouth and are distributed from Eastern and Southern Turkey, Syria, Northern Iraq to Western Iran. The main aim of present study is to determine the phylogenetic placement of S. sect. Satureoides based on Stachys melampyroides, as the only representative of this section in Iran. The present molecular study was conducted based on nuclear ribosomal ITS marker, using Maximum Parsimony and Bayesian Inference analyses. In total, 86 accessions (including nine outgroups and 77 ingroups) were included. Our results demonstrated that sect. Satureoides was placed in Olisia clade which is one of the most heterogenous and taxonomically challenging clades of Stachys as a paraphyletic genus. Moreover, not only sect. Satureoides was not found to be phylogenetically close to sect. Neurocalyx, as had been suggested by previous morphological studies, but also it was placed as a sister group of sect Fragilicaulis which was characterized by chasmophilus species with fragile stems and yellow flowers. Our results revealed that sect. Neurocalyx could be the closest group to the Satureoides+Fragilicaulis clade. In addition, based on the nrITS phylogenetic tree, annual species of Stachys showed affinity to perennial Stachys species. Hence, it seems that “Annual habit” have been evolved among their perennial ancestors, independently.
