fa بررسی همبستگی پلی مورفیسم تک نوکلئوتید rs11614913 ژن mir196a2 با استعداد ابتلا به سرطان پستان در جمعیت جنوب ایران Evaluation of single nucleotide polymorphism rs11614913 in mir196a2 with breast cancer susceptibility in Southern Iranian population مقاله پژوهشی Original Article سرطان پستان شایع ترین سرطان در دنیا و دومین علت مرگ ومیر پس از سرطان ریه در زنان می باشد. میزان شیوع سرطان پستان در کشورهای آسیایی مانند ایران رو بهافزایش است. در طول چند سال گذشته نقش بسیاری از میکروRNAها (miRNA) در بروز انواع سرطان ها مشخص شده است. چندشکلی تکنوکلئوتیدی (SNP) در طول توالی miRNA در ارتباط با خطر ابتلا به انواع سرطانها ازجمله سرطان پستان بررسی شده است. هدف این مطالعه بررسی ارتباط میان چندشکلی تکنوکلئوتیدی rs11614913 در mir196a2 و استعداد ابتلا به سرطان پستان می‌باشد. این مطالعه به بررسی rs11614913 در hsa-mir196a2 در 100 مورد مبتلا به سرطان پستان و 100 مورد کنترل در جمعیت زنان جنوب ایران می پردازد. روش مورد استفاده برای تعیین این چندشکلی، روش RFLP- PCR بود. محصول PCR تحت اثر آنزیم محدودکنندۀ TaaI قرار گرفت و باندهایی به طول 187 و 197 جفت باز مشاهده شد. این مطالعۀ مورد- شاهدی به وسیلۀ نسبت شانس (ORs) با فاصلۀ اطمینان 95٪ (CI) برای آشکار ساختن ارتباط این SNP در mir196a2 با استعداد ابتلا به سرطان پستان تحلیل گردید. فراوانی الل C در mir196a2 در گروه بیماران 0/74و در گروه کنترل 0/75 و فراوانی ژنوتیپی TT،TC وCC  به ترتیب 0/05، 0/42 و 0/53 در گروه بیمار و در گروه کنترل 0/06، 0/38 و 0/56 برآورد شد. در ضمن بررسی آماری نشان دهندۀ وجود تعادل هاردی- واینبرگ بین دو الل این ژن در جمعیت مورد مطالعه بود. باتوجه به مطالعۀ انجام شده مشخص شد هیچگونه ارتباط معناداری میان این پلی مورفیسم و استعداد ابتلا به سرطان پستان وجود ندارد. Breast cancer is the most common cancer worldwide and is the second leading cause of death in women after lung cancer. Micro RNAs (miRNAs) are among endogenous factors which are involved in many types of cancers, including breast cancer. Single nucleotide polymorphism (SNP) in the miRNAs, might change their biological activities such as their effects on oncogenes and tumor suppressor genes. Therefore some of miRNA’s SNPs are associated with the risk of different types of cancer, including breast cancer. The aim of this study was to evaluate the correlation between SNP rs11614913 in mir196a2 and the risk of breast cancer. SNP rs11614913 in hsa-miR-196a2 analyzed in 100 breast cancer cases and 100 controls in women living in southern Iran. Polymorphism was identified by the PCR-RFLP method. The PCR product was digested with TaaI restriction enzyme which produced two bands with the length of 187bp and 196bp. The data from this case –control study were analyzed using odds ratios (ORs) with 95% confidence intervals (CIs) to reveal the associations of SNPs in miRNAs with breast cancer susceptibility. The C allele frequencies in patients and controls for miR-196a2 were 0.74 and 0.75, respectively. The genotype frequencies of TT, TC, and CC were 0.05, 0.42, and 0.53 for the patients and 0.06, 0.38, and 0.56 for the controls, respectively. Statistical analysis showed that the genetic frequencies for this SNP were in Hardy-Weinberg equilibrium (p>0.05).
