Abstract

Saponins are secondary metabolites that are found in many plants and some animals. These compounds are high molecular weight glycosides, consisting of a sugar moiety linked to a triterpene or steroid aglycone. Many saponins have detergency properties and give stable foam in water. The genus Silene L. with more than 700 species is one of the largest genera of the family Caryophyllaceae mainly distributed in northern hemisphere. Saponins are one of the important secondary metabolites in the members of the genus. Quantitative and qualitative study of the saponins in the aerial parts and roots of three Silene species, namely S. ginodioica Ghaz. subsp. Penducularis (Fenzl ex Boiss.) Melzh., S. spergulifolia (Willd.) M. Bieb. and S. swertiifolia Boiss. were carried out by spectrophotometry and TLC methods. In this study, 5 fractions were obtained from each plant part and in both parts of the three species, fraction 1 contains the highest amounts of saponins. On the other hands, root extracts have high amounts of saponins. In addition, thin layer chromatography (TLC) of the extracts revealed some saponins spots with different Rf.

Abstract

Trigonella L. is one of the important genera of the family Fabaceae, many species of which have nutritional and medicinal properties. In this investigation, morphological characters related to habit and pollen grain of three Trigonella species i.e., T. disperma, T. subenervis and T. aphanoneura were studied. Oils from the aerial parts of two species were investigated by GC and GC/MS methods and compared with T. disperma, which had been studied in advance. T. subenervis represented oils rich in spathulenol (15.1%). It also contained a small amount of germacrene D (0.6%). T. aphanoneura also represented oils rich in spathulenol (10.4%). The amount of four compounds in this species was ≤1%, and n-octyl acetate was the lowest component in quantity. According what we found out, phytochemical and morphological data could clarify the relationships among the Trigonellaspecies better than pollen morphological data

Abstract

Alpha-amylases are the most important amylases used in industry. Among them, thermophilic alpha-amylases are of particular importance, which is due to their activity and stability in high temperatures. These enzymes produced by thermophile micro-organisms including bacteria. These thermophilic alpha-amylases are used in various industries such as processing of starch as well as production of detergents and biofuels. In this research, the bacteria which produce the thermophilic alpha-amylases were isolated and characterized in hot springs of Gorooh village in Kerman province. According to the results of screening on the specific liquid and solid media, AT59 was selected as the best strain. Morphological and biochemical characterization of the isolated strain indicated that it belonged to Bacillus sp. and was gram-positive, catalase positive, casein hydrolyzing and acid producing from lactose and sucrose. The results obtained from the optimization of the enzyme production medium showed that among the carbon, nitrogen and ion sources investigated, starch (1 gr/l), gelatin (2 g/l) and magnesium sulfate (1 g/l) had the most increasing effect on the production of AT59 alpha-amylase. Moreover, the highest enzyme production was obtained at pH 5. This enzyme also demonstrated the highest degree of activity and stability in 80 and 70 ℃, respectively. These findings suggested that this enzyme has a considerable potential for use in starch industry.

Abstract

This study aimed to investigate the protective effect of Arctium lappa (AL) on gentamicin (GM)-induced nephrotoxicity in rats. Twenty-four Wistar rats were divided into four groups including: control group; GM group (intrapritoneal injection, IP, of 100 mg/kg GM B.W.); GM+AL group (received IP injection of 100 mg/kg GM and 500 mg/kg AL orally) and AL group (received 500 mg/kg AL orally). The experimental period lasted for 10 days. Nephrotoxicity was biochemically and histologically evaluated. The concentrations of creatinine, urea, malondialdehyde (MDA), superoxide dismutase (SOD) and peroxide hydrogen (H2O2) in the serum samples were determined. Moreover, histological examinations were performed. The animals treated with gentamicin showed significantly higher serum urea, creatinine, MDA and H2O2 levels and lower SOD activity. However, co-administration of AL produced amelioration in biochemical indices of nephrotoxicity in serum. Histomorphological examination showed necrosis and desquamation of tubular epithelial cells in the renal cortex in animals treated with gentamicin whereas simultaneous administration of AL and GM reduced histological damages. The data obtained suggest that treatment with AL extract can help to reduce gentamicin-induced nephrotoxicity.

Polyploidy induction in German chamomile (Matricaria chamomilla L.) by herbicide trifluralin

Abstract. Chamomile (Matricaria chamomilla L.) is a medicinal herb belonging to Asteraceae family. This study was conducted to investigate the effect of trifluralin on the induction of polyploidy in chamomile. Two independent full factorial experiments on seedlings and 2-leaf stage apical buds were performed.  Following morphological, biochemical and cytogenetic analysis, chromosome numbers of 18 and 36 were detected for diploid and tetraploid types, respectively. Morphological and biochemical examinations revealed that an increase in the number of full chromosome set results in a reduction in stomata number per unit area as well as an increase in stomata size, chloroplast number, and chlorophyll content. Induction of ploidy level increments also reduced plant height and increased the number of lateral branches, leaf size, and diameters of stems, flowers and receptacles. It is concluded that a concentration of 22.5 µM trifluralin in both methods is optimum for the production of tetraploid chamomile with the highest rate of polyploidy induction and the lowest percentage of abnormality.

Abstract

Distribution of rivers, pools and grasslands, being large or small and perennial or temporary, have created conditions suitable for the establishment of wetlands, especially in the highland regions of the Country. These habitats are highly important for the preservation of biodiversity and other ecological aspects. The present study aimed to identify wetlands in the Kermanshah Province, Iran, and map their distribution. The investigation was carried out on the basis of the conventional taxonomic studies of the region by doing field observation in 31 aqueous habitats in Kermanshah Province. The flora of each habitats and the life form and geographical distribution of the species were investigated. In total, 617 plant specimens, belonging to 288 species, 218 genera and 62 families, were collected identified and stored in the herbarium of the research center of agriculture and natural resources of Kermanshah Province (RANK). Asteraceae with 41 species, Poaceae with 31 species, Fabaceae with 31 species and Brassicaceae with 24 species were found to be the most important families of the wetlands studied.  Trifolium with 11 species and Bromus with 6 species were the most important genera, in terms of species richness. Detection of the biological forms showed that 40.3% of the species were trophytes, 35/5% were hemicryptophyts, 6/1% were phanerophytes, 14/23% were cryptophytes and 3/72% were chamaephytes. The chorology of species showed that 93 species belonged to more than two elements (32%), 92 species (31%) to the Irano-Touranin elements, 32 species to Irano-Touranin- Mediterranean (11%), 9 species to Irano-Touranin- Euro-Siberian with (3%) and 23 species were found to be cosmopolitan elements (8%).

Abstract

It has been found that the second isoform of COX enzyme known as COX-2 plays an important role in inflammation and rheumatoid arthritis and osteoarthritis. Thus, designing COX-2 inhibitors to treat inflammation is among the most important goals of researchers. In this study, the inhibitory effect of 3 new imidazole derivatives on COX-2 was evaluated by in silico approach. Molecular docking was done using Autodock Vina and the best binding mode of inhibitors was used as input of molecular dynamics (MD) simulation. MD was performed using Gromacs software for 120 ns. Then, structural and thermodynamic analyzes (ΔGbinding) and prediction of physicochemical properties were performed. RMSD data showed the compounds reached a good equilibrium and had favorable stability during simulation. Also, the RMSF showed that due to binding of inhibitors, the fluctuations of complexes decreased and the active site residues had the lowest amount. Rg, SASA and DSSP analysis showed that the protein structure did not change significantly. It was also found that Ser530 and Tyr355 residues play a more effective role in hydrogen bond formation. Physicochemical parameters determined the good drug-likeness properties for all compounds. Structural and thermodynamic analyzes (MM-PBSA) and IC50 data indicate the favorable inhibitory effect of compound 5b.
